PROJECT TITLE
“Regenerative Mechanisms for Health" ReMedy
SUBPROJECT TITLE:
“Bunyaviral translation strategies”
PROJECT LEADER
Head: Piotr Gerlach / Laboratory of Structural Virology
SUBPROJECT DESCRIPTION
Host translation is one of the major sites of the virus-host battlefront and a center of cellular stress response pathways triggered during viral infection. The aim of this project is to investigate how segmented negative-strand RNA bunyaviruses manipulate and reprogramme cellular host translation during infection.
Methodologically we plan to integrate structural biology techniques (cryo-EM, cryo-ET, and X-ray crystallography) with biochemistry and in vivo approaches. Our lab has access to state-of-the-art instrumentation, including cell culture facilities, recombinant protein purification systems, mass spectrometry, and a Glacios cryo-TEM equipped with Falcon 3EC camera. Collaborations within the Ochota Life Science campus afford access to two FIB-SEM instruments for cryo-ET sample preparation. Finally, we will have access to a Titan Krios G3i cryo-TEM equipped with both K3 and Falcon3EC cameras, located at the National Centre for Electron Cryomicroscopy in Kraków.
The successful candidate will be involved in setting up an in vivo platform – mammalian cell cultures transfected with viral mini-replicon systems, which will be exploited in functional and structural characterization of the virus-host intracellular interactions. Experience in mammalian cell cultures, including purification of native protein complexes, is desired. Experience in cryo-EM/ET sample preparation and data processing would be an advantage.
